Main duties and responsibilities
You’ll have the opportunity to identify mechanisms by which oncotherapeutics affect adult intestinal stem cells, progenitor cells, and differentiating enterocytes. Your work will utilize cutting-edge molecular and cellular biology techniques to dissect essential elements of stem cell self-renewal and maturation in the intestine. In addition to expanding frontiers in stem cell biology, your work will directly contribute to understanding the adverse mechanisms that are presently prohibitive for entirely new classes of oncotherapeutics.

You will be part of a post doc program that aims to offer exposure to drug discovery science and careers while maintaining your ability to publish. While you will have the opportunity for collaborative support from scientists working on diverse oncology drug discovery projects, your work will focus exclusively on discovering novel mechanism limiting cancer therapy and publication thereof.

Key tasks will be to define intestinal stem cell and differentiating enterocyte populations using in vitro, in vivo, and in silico methodologies and quantify their sensitivity to various pharmacological agents. You will use bioinformatics to identify key transcriptional regulators of intestinal stem cell self-renewal and proliferation. You’ll test the essentiality of the identified genes in cutting-edge organoid systems and identify their key downstream targets, and verify your findings in vivo using transgenic animal systems. We’ll encourage you to publish your findings in high-impact, peer-reviewed scientific journals. Working collaboratively with experts internally and externally across various disciplines you’ll also present results externally at scientific conferences and meetings.

Essential requirements
- PhD and/or MD in relevant discipline, such as molecular biology, physiology, pharmacology or toxicology
- Expertise with standard molecular biology techniques, including mammalian cell culture
- Proven scientific track record of publications in reputable scientific journals
- Commitment to medical sciences

Desirable requirements
- Previous experience with culturing stem cells and/or organoid systems; application of these systems to study intestinal biology is a particular advantage
- Experience with flow cytometry and bioinformatics
- Knowledge of cell signaling pathways
- Experience working with lab animals

This is a 2 year Fixed Term Contract opportunity, plus 1 year which will be merit based. The role will be based at Waltham, MA, US with a competitive salary on offer. The projects will employ mouse models, organoid cultures, and epigenomic (ChIP-seq) methodologies. Postdocs are encouraged to take any developed projects with them as their careers progress.

Job Description
Vertex Pharmaceuticals is looking for an outstanding Cell Biologist to join our drug discovery team in San Diego. This position will focus on applying and developing novel human cell biology and tissue engineering technologies to support discovery and development of transformative medicines. The successful candidate should have a strong scientific background and proven record of accomplishments in the areas of primary human stem cell biology using 3D scaffolds and relevant biomaterials. The ideal candidate will be responsible for generation, maintenance, differentiation and characterization of primary human progenitors, stem cells and organoids. Extensive hands-on experience in working with primary human cells, developing complex functional assays, high content imaging, and implementing quantitative image analysis using commercial and open-source software is required. The applicant will be part of a fast-paced drug discovery team that includes cell biologists, molecular biologists, pharmacologists, and engineers. In addition, the position will closely interact with an Instrumentation and Biophysics group with capabilities in traditional engineering, micro-fabrication, microfluidics, biomaterial scaffolds, and applied physics.
Minimum Qualifications

- Ph.D. in Biology, Bioengineering or equivalent and minimum of 5 years of postgraduate experience in tissue engineering & primary human cell biology - He/She should have a deep understanding of 3D scaffolds and the use of biomaterial for the growth of human cells - Proven track record in setting up physiologically relevant cell based assays using primary human cells, 3D cultures and organoids is required - Proficient with confocal microscopy, high content image acquisition, implementation of quantitative image analysis using state of the art imaging software (such as MetaMorph, Volocity, Cell Profiler) is essential - Experience in drug discovery in biotech/pharmaceutical setting is required - Proficiency in various cell biology and molecular biology technical skills is required - Excellent communication skills and adaptive to a time-line driven work environment - Exceptional multitasking skills, attention to details and ability to work independently are required - Results orientated, driven, motivated, and well organized Preferred Qualifications - Basic understanding of drug discovery process is preferred - Ability to navigate and be successful in a fast-paced, highly-matrixed work environment - A desire to be part of a highly innovative company aimed at transforming the lives of people with serious diseases, their families and society

2015 will be the year of the AbbVie Postdoctoral Program launch. We are looking for the incoming class to join AbbVie as the company is positioning itself for another significant year of growth and of the continuing advancement of its patient-driven science and its strategic priorities. In a collaborative, resource rich environment, you and your scientific mentor will be exploring, targeting, investigating and evaluating key areas of novel research with a publication as the end point. We have an exceptional postdoctoral opportunity within our Foundational Immunology Group reporting into a Principal Scientist studying mucosal immunology at the AbbVie Bioresearch Center (ABC) based in Worcester, Massachusetts.

ABC is a fully integrated biologics research and development and commercial manufacturing site with activities spanning the discovery, development, commercial launch and product life cycle management of the AbbVie biologics pipeline. The Foundational Immunology Group is seeking a motivated and innovative applicant to the postdoctoral program to investigate mechanisms of intestinal epithelial cell function using human intestinal organoids. This research program is critical to the broader Immunology strategy and is expected to deliver new insights into biology relevant to inflammatory bowel disease (IBD) and assist with identification and validation of drug discovery programs that will form the basis of next generation therapeutics for IBD.  

 

Key responsibilities:

  • Establish 3-D and 2-D human intestinal organoid culture systems
  • Develop methodology to study specific epithelial cell functions (i.e. tight junction formation, mucus production, defensin production, barrier function)
  • Investigate candidate epithelial biomarkers associated with human IBD
  • Establish coculture conditions with myofibroblasts to study their influence on organoid behavior
  • Establish protocols for genetic modification of organoids by retroviral overexpression/knockdown or with CRISPR technology to understand the effect such modifications have on organoid growth, differentiation, survival, and function
  • Develop wound assays to explore processes involved with healing

Basic qualifications:

  • PhD  in Immunology, Molecular or Cellular Biology, or a related discipline from an accredited and nationally ranked university
  • Background in epithelial cell biology and organoid culture development
  • Experience with immune cell biology and primary culture of immune cells
  • Excellent in vitro and molecular biology skills
  • Familiarity with the pathophysiology of IBD
  • Authorized to work in the U.S.
  • Provide a narrative on why your academic training would qualify you for this position  and why you have a sustained interest in this program
  • Provide three letters of recommendation

Preferred qualifications:

  • Experience with mammalian cell signaling research
  • Experience with myofibroblast biology and culturing
  • Experience with genetic manipulation of organoids
  • Record of publication in a well-recognized journal(s)
  • Ability to effectively work both independently and collaboratively
  • Excellent problem-solving, communication, and project management skills
